Choosing the Best Leukopak Option for Your Research

Collection lead times are typically two weeks for non-mobilized leukapheresis collections and six weeks for mobilized leukapheresis collections; lead times may vary depending on specificity of requested donor characteristics and collection criteria. With iSpecimen, you can also select for specific mobilization products such as G-CSF filgrastim, Mobizil (plerixafor), or a combination of these products. Leukopaks can be shipped under cold or ambient conditions as requested, with an option to include a temperature logger.

Fresh Healthy Mobilized Leukopak Collection

Mobilized for Maximum Cell Yield

To stimulate egress of hematopoietic stem cells (HSCs) from the bone marrow into peripheral blood, healthy consented donors are mobilized using granulocyte colony-stimulating factor (G-CSF, filgrastim/Neupogen) administered via subcutaneous injection on a daily basis. Depending on the specific mobilization protocol desired, healthy donors can be mobilized daily for one (1) to five (5) days followed by a single mononuclear cell collection plus an option for a second-day collection to maximize total cell yield from a single donor. For example, Day 1 Leukopak collection may typically yield >50 billion cells and Day 2 collections may typically yield >30 billion cells in a full Leukopak, subject to donor variability.

Cryopreserved Diagnosed Mobilized Leukopaks (Cancer)

iSpecimen offers a banked inventory of cryopreserved mobilized leukopaks collected from early-stage cancer patients who have been mobilized using a 5-day GRANIX® (tbo-filgrastim) stimulation regimen. leukopaks are available from patients diagnosed with ALL, CLL, MM, CML, Follicular Lymphoma, prostate and ovarian cancer, and renal failure. Each leukopak is rapidly cryopreserved and stored in the liquid nitrogen vapor phase at <-150 Deg. C to maintain optimal quality. Each leukopak varies in volume, total nucleated cell count, cellularity, and percent cell viability depending on the collection conditions used for each patient donor.

Cryopreserved leukopaks may be obtained from patients diagnosed with Crohn’s Disease, Psoriasis, Celiac Disease, Ulcerative Colitis, Lupus, Sjogren’s Syndrome, Diabetes, and others.

(Non-Mobilized) Leukopaks

Fresh Healthy Normal, Non-Mobilized Leukopak Collection

Mononuclear cells are collected and concentrated from peripheral blood of healthy consented donors according to IRB approved protocols using an automated leukapheresis system. During the procedure, an anticoagulant is added to venous blood to prevent blood from clotting as it passes through the apheresis device.

The enriched cell fraction consisting of pan T, NK, B cells, hematopoietic stem cells (CD34+), monocytes, dendritic, and other white blood cell types is collected and shipped in a sterile blood bag. Collection volumes for a full non-mobilized fresh Leukopak range from 300-500 mL and cell enumeration is typically >10 billion cells with cell yields as high as 28 billion, subject to donor variability.

Cryopreserved Healthy Normal (Non-Mobilized) Leukopak Collection

Freshly collected mononuclear cells are carefully cryopreserved using a 10% DMSO cryoprotectant in a sterile CRYO bag. Each leukopak is cryopreserved at a US-FDA registered, AABB accredited facility using a controlled rate freezer according to a validated processing protocol. Each leukopak is stored in the liquid nitrogen vapor phase at <-150 Deg. C to maintain optimal product quality. Cryopreserved leukopaks are ready to be thawed and may allow for additional flexibility in conducting R&D studies depending on your changing schedule.
